Ronaldo, who was crowned UEFA Best Player in Europe on Thursday, will be rested after admitting he was not at full fitness.
Head coach Carlo Ancelotti said: "We have decided to give some days' rest to Cristiano He will not be part of the squad for tomorrow."
Ronaldo has played all four of Real’s matches this month – the UEFA Super Cup, both legs of the Spanish Super Cup against Atletico Madrid and last weekend’s Liga opener against Cordoba.
Ronaldo has been battling a hamstring strain, and said he played at the World Cup for Portugal while still suffering from a knee injury.
He told Marca on Friday: "We were involved in important competitions with the Champions League and then the World Cup.
"Then there were ups and downs in pre-season which is normal with the Real Madrid tour, the trips, the tiredness and jet-lag. All this means that you can't work well.
"Last week I was running okay but I still need some days to be at my best although it will be soon.
"Clearly I did (force myself too much). I should probably have stopped and now I would be fine."
